Numb is a transmembrane signaling receptor that is activated by the DSL family of ligands. Studies suggest that mammalian Numb plays a role in progenitor cell fate in neurogenesis. Numb exerts its role on cell fate by antagonizing Notch signaling. The mammalian Numb gene undergoes alternative splicing to produce at least four functionally different Numb isoforms which regulate different processes. A loss of Numb expression has been demonstrated in breast cancer, non-small cell lung cancer and chronic myelogenous leukemia.
