Background :
PDK1 (pyruvate dehydrogenase kinase isoform 1) belongs to PDK/BCKDK protein kinase family and is a serine/threonine kinase which regulates glucose and fatty acid metabolism/homeostasis via phosphorylation of pyruvate dehydrogenase (PDH) subunits PDHA1 and PDHA2. PDK1 is mainly expressed in cardiac tissue and it localizes to mitochondrion matrix where it phosphoryles E1 alpha subunit of mitochondrial PDH complex, leading to PDH inhibition thus contributing to the regulation of glucose metabolism. PDK1 inhibits PDH activity, and thus regulates metabolite flux through the TCA/Kreb's cycle, down-regulates aerobic respiration and inhibits the formation of acetyl-coenzyme A from pyruvate. Because, PDH is a gatekeeper enzyme that converts pyruvate to acetyl-CoA, PDK1 is also regarded as a key regulator of the Warburg effect. PDK1 plays an important role in cellular responses to hypoxia and is important for cell proliferation under hypoxia. Protects cells against apoptosis in response to hypoxia and oxidative stress. It is inhibited by AZD7545, dichloroacetate and radicicol and these inhibitors have been tested for anticancer activity. Knockdown of PDK1 by shRNA lowers PDH phosphorylation, reactivates PDC activity and decreases invasiveness in human head and neck squamous cell carcinoma.

This PDK1 antibody is useful for Immunocytochemistry/Immunofluorescence and Western Blot. By Western blot a band is seen at ~49 kDa, representing PDK1 protein. There are also two higher and one lower non-specific molecular weight bands. In ICC/IF, mitochondiral staining was observed in HeLa cells.
