Background :
Originally identified as an AR -interacting protein with ability to inhibit AR-mediated transcriptional responses, PAK6 (p21-activated kinase 6) is now known as a serine/threonine protein kinase implicated in regulation of gene transcription. It belongs to PAK family which has two groups - Group I (PAK1, PAK2 and PAK3) and Group II PAKs (PAK4, PAK5 and PAK6), and PAK members are critical in the regulation of multiple processes including actin reorganization, cell motility, gene transcription, cell transformation, apoptotic signaling, and steroid hormone receptor signaling. PAK6 is selectively expressed in brain and testis with low levels in other tissues such as prostate, breast etc, and it interacts with several proteins that include GTP-bound CDC42/p21, RAC1, AR, ESR1, IQGAP1 and PPM1B. MAP2K6/MAPKK6 and AR can induce PAK6's kinase activity and it phosphorylate DNA-binding domain of AR leading to inhibition of AR-mediated transcription. Moreover, PAK6 suppresses prostate carcinogenesis by impairing AR nuclear translocation and by promoting recruitment of E3 ligase Mdm2 to AR for its degradation via ubiquitinization. PAK6 provides protection against apoptosis via BAD phosphorylation, inhibits ESR1-mediated transcription and involves in cytoskeleton regulation by interacting with IQGAP1.
