Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for CA1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CA1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CA1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CA1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Carbonic anhydrases (CAs) are a large family of zinc metalloenzymes that catalyze the reversible hydration of carbon dioxide. They participate in a variety of biological processes, including respiration, calcification, acid-base balance, bone resorption, and the formation of aqueous humor, cerebrospinal fluid, saliva and gastric acid. They show extensive diversity in tissue distribution and in their subcellular localization. This CA1 gene is closely linked to the CA2 and CA3 genes on chromosome 8. It encodes a cytosolic protein that is found at the highest level in erythrocytes. Allelic variants of this gene have been described in some populations.
