Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for FCGR3A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any FCGR3A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for FCGR3A is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FCGR3A bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a receptor for the Fc portion of immunoglobulin G, and it is involved in the removal of antigen-antibody complexes from the circulation, as well as other other antibody-dependent responses. This gene (FCGR3A) is highly similar to another nearby gene (FCGR3B) located on chromosome 1. The receptor encoded by this gene is expressed on natural killer (NK) cells as an integral membrane glycoprotein anchored through a transmembrane peptide, whereas FCGR3B is expressed on polymorphonuclear neutrophils (PMN) where the receptor is anchored through a phosphatidylinositol (PI) linkage. Mutations in this gene have been linked to susceptibility to recurrent viral infections, susceptibility to systemic lupus erythematosus, and alloimmune neonatal neutropenia.
