Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for TGM1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TGM1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TGM1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TGM1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is a membrane protein that catalyzes the addition of an alkyl group from an akylamine to a glutamine residue of a protein, forming an alkylglutamine in the protein. This protein alkylation leads to crosslinking of proteins and catenation of polyamines to proteins. This gene contains either one or two copies of a 22 nt repeat unit in its 3' UTR. Mutations in this gene have been associated with autosomal recessive lamellar ichthyosis (LI) and nonbullous congenital ichthyosiform erythroderma (NCIE). [provided by RefSeq, Jul 2008]
