Principle of the Assay This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ABL1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ABL1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ABL1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ABL1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene is a protooncogene that encodes a protein tyrosine kinase involved in a variety of cellular processes, including cell division, adhesion, differentiation, and response to stress. The activity of the protein is negatively regulated by its SH3 domain, whereby deletion of the region encoding this domain results in an oncogene. The ubiquitously expressed protein has DNA-binding activity that is regulated by CDC2-mediated phosphorylation, suggesting a cell cycle function. This gene has been found fused to a variety of translocation partner genes in various leukemias, most notably the t(9;22) translocation that results in a fusion with the 5' end of the breakpoint cluster region gene (BCR; MIM:151410).
