specificity :
This assay has high sensitivity and excellent specificity for detection of human PAWR. No significant cross-reactivity or interference between human PAWR and analogues was observed.
storage stability :
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
other info1 :
Samples: Serum, plasma, tissue homogenates, Cell lysates. Assay Type: Sandwich. Detection Range: 25 pg/ml -1600 pg/ml. Sensitivity: The minimum detectable dose of human PAWR is typically less than 6.25 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
products description :
Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for PAWR has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PAWR present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PAWR is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PAWR bound in the initial step. The color development is stopped and the intensity of the color is measured.

ncbi summary :
The tumor suppressor WT1 represses and activates transcription. The protein encoded by this gene is a WT1-interacting protein that itself functions as a transcriptional repressor. It contains a putative leucine zipper domain which interacts with the zinc finger DNA binding domain of WT1. This protein is specifically upregulated during apoptosis of prostate cells. uniprot summary :
PAR-4: a proapoptotic protein that drives trafficking and activation of Fas and Fasl to induce prostate cancer cell apoptosis and tumor regression. Capable of selectively inducing apoptosis in cancer cells, sensitizing the cells to diverse apoptotic stimuli and causing regression of tumors in animal models. Induces apoptosis in certain cancer cells by activation of the Fas prodeath pathway and coparallel inhibition of NF-kappaB transcriptional activity. Inhibits the transcriptional activation and augments the transcriptional repression mediated by WT1. Down-regulates the anti-apoptotic protein BCL2 via its interaction with WT1. Seems also to be a transcriptional repressor by itself. May be directly involved in regulating the amyloid precursor protein (APP) cleavage activity of BACE1. Protein type: Apoptosis. Chromosomal Location of Human Ortholog: 12q21. Cellular Component: cytoplasm; plasma membrane; actin filament; nucleus; actin cytoskeleton. Molecular Function: protein binding; leucine zipper domain binding; enzyme binding; actin binding; transcription corepressor activity. Biological Process: actin filament bundle formation; negative regulation of cell proliferation; interleukin-2 biosynthetic process; negative regulation of T cell proliferation; transcription, DNA-dependent; apoptosis; positive regulation of apoptosis; negative regulation of T cell receptor signaling pathway; negative regulation of transcription from RNA polymerase II promoter; negative regulation of B cell proliferation; positive regulation of amyloid precursor protein biosynthetic process
