specificity :
This assay has high sensitivity and excellent specificity for detection of rat ROCK2. No significant cross-reactivity or interference between rat ROCK2 and analogues was observed.
storage stability :
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
other info1 :
Samples: Serum, plasma, tissue homogenates. Assay Type: Sandwich. Detection Range: 62.5 pg/ml -4000 pg/ml. Sensitivity: The minimum detectable dose of rat ROCK2 is typically less than 15.6 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
products description :
. Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ROCK2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ROCK2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ROCK2 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ROCK2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

ncbi summary :
a Rho kinase that is involved in inducing membrane blebbing and chromatin condensation [RGD, Feb 2006]
uniprot summary :
ROCK2: Protein kinase which is a key regulator of actin cytoskeleton and cell polarity. Involved in regulation of smooth muscle contraction, actin cytoskeleton organization, stress fiber and focal adhesion formation, neurite retraction, cell adhesion and motility via phosphorylation of ADD1, BRCA2, CNN1, EZR, DPYSL2, EP300, MSN, MYL9/MLC2, NPM1, RDX, PPP1R12A and VIM. Phosphorylates SORL1 and IRF4. Acts as a negative regulator of VEGF-induced angiogenic endothelial cell activation. Positively regulates the activation of p42/MAPK1-p44/MAPK3 and of p90RSK/RPS6KA1 during myogenic differentiation. Plays an important role in the timely initiation of centrosome duplication. Inhibits keratinocyte terminal differentiation. May regulate closure of the eyelids and ventral body wall through organization of actomyosin bundles. Plays a critical role in the regulation of spine and synaptic properties in the hippocampus. Homodimer. Interacts with IRS1, RHOB and RHOC. Interacts with RHOA (activated by GTP), PPP1R12A, CHORDC1, SORL1, EP300 and BRCA2. Interacts with NPM1 and this interaction enhances its activity. Interacts with RAF1. Activated by RHOA binding. Inhibited by Y-27632. Belongs to the protein kinase superfamily. AGC Ser/Thr protein kinase family. Protein type: Protein kinase, AGC; Kinase, protein; EC 2.7.11.1; Protein kinase, Ser/Thr (non-receptor); AGC group; DMPK family; ROCK subfamily. Cellular Component: centrosome; cytoplasm; plasma membrane; microtubule organizing center; spindle pole centrosome; nucleus. Molecular Function: protein serine/threonine kinase activity; protein binding; Rho GTPase binding; metal ion binding; kinase activity; ATP binding. Biological Process: dendrite morphogenesis; rhythmic process; regulation of circadian rhythm; protein amino acid phosphorylation; Rho protein signal transduction; chromosome condensation; negative regulation of angiogenesis; smooth muscle contraction; induction of apoptosis via death domain receptors; regulation of actin cytoskeleton organization and biogenesis; neural tube closure; actin cytoskeleton organization and biogenesis; regulation of keratinocyte differentiation; centrosome duplication
