Principle of the Assay This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for SPINK5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SPINK5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SPINK5 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SPINK5 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The inhibitor may play a role in skin and hair morphogenesis and anti-inflammatory and/or antimicrobial protection of mucous epithelia. Mutations may result in Netherton syndrome, a disorder characterized by ichthyosis, defective cornification, and atopy. NETH is an autosomal recessive congenital ichthyosis associated with hair shaft abnormalities and anomalies of the immune system. Typical features are ichthyosis linearis circumflexa, ichthyosiform erythroderma, trichorrhexis invaginata (bamboo hair), atopic dermatitis, and hayfever. High postnatal mortality is due to failure to thrive, infections and hypernatremic dehydration. 3 isoforms of the human protein are produced by alternative splicing.
