Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for DEFA5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any DEFA5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DEFA5 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DEFA5 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Defensins are a family of microbicidal and cytotoxic peptides thought to be involved in host defense. They are abundant in the granules of neutrophils and also found in the epithelia of mucosal surfaces such as those of the intestine, respiratory tract, urinary tract, and vagina. Members of the defensin family are highly similar in protein sequence and distinguished by a conserved cysteine motif. Several of the alpha defensin genes appear to be clustered on chromosome 8.
