Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for PGK1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PGK1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PGK1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PGK1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is a glycolytic enzyme that catalyzes the conversion of 1,3-diphosphoglycerate to 3-phosphoglycerate. The encoded protein may also act as a cofactor for polymerase alpha. Additionally, this protein is secreted by tumor cells where it participates in angiogenesis by functioning to reduce disulfide bonds in the serine protease, plasmin, which consequently leads to the release of the tumor blood vessel inhibitor angiostatin. The encoded protein has been identified as a moonlighting protein based on its ability to perform mechanistically distinct functions. Deficiency of the enzyme is associated with a wide range of clinical phenotypes hemolytic anemia and neurological impairment.
