Principle of the Assay This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for CST1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CST1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CST1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CST1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The cystatin superfamily encompasses proteins that contain multiple cystatin-like sequences. Some of the members are active cysteine protease inhibitors, while others have lost or perhaps never acquired this inhibitory activity. There are three inhibitory families in the superfamily, including the type 1 cystatins (stefins), type 2 cystatins and the kininogens. The type 2 cystatin proteins are a class of cysteine proteinase inhibitors found in a variety of human fluids and secretions, where they appear to provide protective functions. The cystatin locus on chromosome 20 contains the majority of the type 2 cystatin genes and pseudogenes.
