ERAP1_HUMAN
specificity :
This assay has high sensitivity and excellent specificity for detection of human ERAP1. No significant cross-reactivity or interference between human ERAP1 and analogues was observed.
storage stability :
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
other info1 :
Samples: Serum, plasma, tissue homogenates. Assay Type: Sandwich. Detection Range: 0.156 ng/ml -10 ng/ml. Sensitivity: The minimum detectable dose of human ERAP1 is typically less than 0.039 ng/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
products description :
Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ERAP1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ERAP1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ERAP1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ERAP1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

ncbi summary :
The protein encoded by this gene is an aminopeptidase involved in trimming HLA class I-binding precursors so that they can be presented on MHC class I molecules. The encoded protein acts as a monomer or as a heterodimer with ERAP2. This protein may also be involved in blood pressure regulation by inactivation of angiotensin II. Three transcript variants encoding two different isoforms have been found for this gene.[provided by RefSeq, Oct 2010]
uniprot summary :
ARTS1: Aminopeptidase that plays a central role in peptide trimming, a step required for the generation of most HLA class I- binding peptides. Peptide trimming is essential to customize longer precursor peptides to fit them to the correct length required for presentation on MHC class I molecules. Strongly prefers substrates 9-16 residues long. Rapidly degrades 13-mer to a 9-mer and then stops. Preferentially hydrolyzes the residue Leu and peptides with a hydrophobic C-terminus, while it has weak activity toward peptides with charged C-terminus. May play a role in the inactivation of peptide hormones. May be involved in the regulation of blood pressure through the inactivation of angiotensin II and/or the generation of bradykinin in the kidney. Belongs to the peptidase M1 family. 2 isoforms of the human protein are produced by alternative splicing. Protein type: EC 3.4.11.-; Protease; Membrane protein, integral. Chromosomal Location of Human Ortholog: 5q15. Cellular Component: extracellular space; endoplasmic reticulum membrane; membrane; endoplasmic reticulum lumen; endoplasmic reticulum; extracellular region; integral to membrane; cytosol. Molecular Function: protein binding; interleukin-6 receptor binding; metalloexopeptidase activity; interleukin-1, Type II receptor binding; zinc ion binding; aminopeptidase activity. Biological Process: fat cell differentiation; positive regulation of angiogenesis; antigen processing and presentation of peptide antigen via MHC class I; response to bacterium; regulation of blood pressure; membrane protein ectodomain proteolysis; angiogenesis; antigen processing and presentation of endogenous peptide antigen via MHC class I; regulation of innate immune response
