Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for RIPK1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any RIPK1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for RIPK1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of RIPK1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Upon activation of TNFR1 by the TNF-alpha family cytokines, TRADD and TRAF2 are recruited to the receptor. Ubiquitination by TRAF2 via Lys-63 -link chains acts as a critical enhancer of communication with downstream signal transducers in the mitogen-activated protein kinase pathway and the NF-kappa-B pathway, which in turn mediate downstream events including the activation of genes encoding inflammatory molecules.
