Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ARMS2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ARMS2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ARMS2 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ARMS2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

ARMS2: Defects in ARMS2 influence susceptibility to age-related macular degeneration type 8 (ARMD8). ARMD is the most common cause of irreversible vision loss in the developed world. In most patients, the disease is manifest as ophthalmoscopically visible yellowish accumulations of protein and lipid (known as drusen) that lie beneath the retinal pigment epithelium and within an elastin-containing structure known as Bruch s membrane. ARMD is likely to be a mechanistically heterogeneous group of disorders, and the specific disease mechanisms that underlie the vast majority of cases are currently unknown.
