Principle of the Assay This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for KISS1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any KISS1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for KISS1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of KISS1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene is a metastasis suppressor gene that suppresses metastases of melanomas and breast carcinomas without affecting tumorigenicity. The encoded protein may inhibit chemotaxis and invasion and thereby attenuate metastasis in malignant melanomas. Studies suggest a putative role in the regulation of events downstream of cell-matrix adhesion, perhaps involving cytoskeletal reorganization. A protein product of this gene, kisspeptin, stimulates gonadotropin-releasing hormone (GnRH)-induced gonadotropin secretion and regulates the pubertal activation of GnRH nuerons. A polymorphism in the terminal exon of this mRNA results in two protein isoforms. An adenosine present at the polymorphic site represents the third position in a stop codon.
