Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for C8B has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any C8B present is bound by the immobilized antibody. After removing any unbound substances, a Horseradish Peroxidase (HRP)-conjugated antibody specific for C8B is added to the wells. Following a wash to remove any unbound reagent, a substrate solution is added to the wells and color develops in proportion to the amount of C8B bound in the initial step. The color development is stopped and the intensity of the color is measured.

C8 is composed of equimolar amounts of alpha, beta and gamma subunits, which are encoded by three separate genes. C8 is one component of the membrane attack complex, which mediates cell lysis, and it initiates membrane penetration of the complex. This protein mediates the interaction of C8 with the C5b-7 membrane attack complex precursor. In humans deficiency of this protein is associated with increased risk of meningococcal infections.
