specificity :
This assay has high sensitivity and excellent specificity for detection of human NANOG. No significant cross-reactivity or interference between human NANOG and analogues was observed.
storage stability :
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
other info1 :
Samples: Serum, plasma, tissue homogenates, Cell lysates. Assay Type: Sandwich. Detection Range: 25 pg/ml -1600 pg/ml. Sensitivity: The minimum detectable dose of human NANOG is typically less than 6.25 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
products description :
Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for NANOG has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any NANOG present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NANOG is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NANOG bound in the initial step. The color development is stopped and the intensity of the color is measured.

ncbi summary :
The protein encoded by this gene is a DNA binding homeobox transcription factor involved in embryonic stem (ES) cell proliferation, renewal, and pluripotency. The encoded protein can block ES cell differentiation and can also autorepress its own expression in differentiating cells. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2015]
uniprot summary :
NANOG: Transcription regulator involved in inner cell mass and embryonic stem (ES) cells proliferation and self-renewal. Imposes pluripotency on ES cells and prevents their differentiation towards extraembryonic endoderm and trophectoderm lineages. Blocks bone morphogenetic protein-induced mesoderm differentiation of ES cells by physically interacting with SMAD1 and interfering with the recruitment of coactivators to the active SMAD transcriptional complexes. Acts as a transcriptional activator or repressor. Binds optimally to the DNA consensus sequence 5 -TAAT[GT][GT]-3 or 5 -[CG][GA][CG]C[GC]ATTAN[GC]-3 . When overexpressed, promotes cells to enter into S phase and proliferation. Interacts with SMAD1 and SALL4. Expressed in testicular carcinoma and derived germ cell tumors. Expressed in fetal gonads, ovary and testis. Also expressed in ovary teratocarcinoma cell line and testicular embryonic carcinoma. Not expressed in many somatic organs and oocytes. Belongs to the Nanog homeobox family. 2 isoforms of the human protein are produced by alternative splicing. Protein type: Transcription factor; DNA-binding; Cell development/differentiation. Chromosomal Location of Human Ortholog: 12p13.31. Cellular Component: nucleoplasm; nucleolus; nucleus. Molecular Function: DNA binding; chromatin binding; transcription corepressor activity; transcription factor activity. Biological Process: gonad development; response to retinoic acid; transcription, DNA-dependent; somatic stem cell maintenance; stem cell division; endodermal cell fate specification; stem cell maintenance; negative regulation of transcription from RNA polymerase II promoter; embryonic pattern specification; positive regulation of mitotic cell cycle; mesodermal cell fate commitment; negative regulation of BMP signaling pathway; negative regulation of cell fate commitment; cell proliferation; regulation of transcription, DNA-dependent; regulation of gene expression; regulation of cell differentiation; positive regulation of cell proliferation; positive regulation of transcription from RNA polymerase II promoter; cell differentiation
