Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for CD81 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CD81 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CD81 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CD81 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Most of these members are cell-surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the regulation of cell development, activation, growth and motility. This encoded protein is a cell surface glycoprotein that is known to complex with integrins. This protein appears to promote muscle cell fusion and support myotube maintenance. Also it may be involved in signal transduction. This gene is localized in the tumor-suppressor gene region and thus it is a candidate gene for malignancies.
