Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for FMR1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any FMR1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for FMR1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FMR1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene binds RNA and is associated with polysomes. The encoded protein may be involved in mRNA trafficking from the nucleus to the cytoplasm. A trinucleotide repeat (CGG) in the 5' UTR is normally found at 6-53 copies, but an expansion to 55-230 repeats is the cause of fragile X syndrome. Expansion of the trinucleotide repeat may also cause one form of premature ovarian failure (POF1). Multiple alternatively spliced transcript variants that encode different protein isoforms and which are located in different cellular locations have been described for this gene.
