Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for PROM1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PROM1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PROM1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PROM1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a pentaspan transmembrane glycoprotein. The protein localizes to membrane protrusions and is often expressed on adult stem cells, where it is thought to function in maintaining stem cell properties by suppressing differentiation. Mutations in this gene have been shown to result in retinitis pigmentosa and Stargardt disease. Expression of this gene is also associated with several types of cancer. This gene is expressed from at least five alternative promoters that are expressed in a tissue-dependent manner.
