This assay has high sensitivity and excellent specificity for detection of human NRTN. No significant cross-reactivity or interference between human NRTN and analogues was observed.

Samples: Serum, plasma, tissue homogenates. Assay Type: Sandwich. Detection Range: 125 pg/ml -8000 pg/ml. Sensitivity: 31.25 pg /ml.

Principle of the Assay This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for NRTN has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any NRTN present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NRTN is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NRTN bound in the initial step. The color development is stopped and the intensity of the color is measured.

Neurturin is a member of the TGF-beta subfamily, TRN. This gene signals through RET and a GPI-linked coreceptor, and promotes survival of neuronal populations. A neurturin mutation has been described in a family with Hirschsprung Disease. [provided by RefSeq, Jul 2008]

NRTN: Supports the survival of sympathetic neurons in culture. May regulate the development and maintenance of the CNS. Might control the size of non-neuronal cell population such as haemopoietic cells. Genetic variations in NRTN may contribute to Hirschsprung disease, in association with mutations of RET gene, and possibly mutations in other loci. Hirschsprung disease is a disorder of neural crest development is characterized by the absence of intramural ganglion cells in the hindgut, often resulting in intestinal obstruction.
