This assay has high sensitivity and excellent specificity for detection of human PEG10. No significant cross-reactivity or interference between human PEG10 and analogues was observed.

Samples: Serum, plasma, tissue homogenates, Cell lysates. Assay Type: Sandwich. Detection Range: 18.75 pg/ml -1200 pg/ml. Sensitivity: The minimum detectable dose of human PEG10 is typically less than 4.69 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.

Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.

Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for PEG10 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PEG10 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PEG10 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PEG10 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This is a paternally expressed imprinted gene that encodes transcripts containing two overlapping open reading frames (ORFs), RF1 and RF1/RF2, as well as retroviral-like slippage and pseudoknot elements, which can induce a -1 nucleotide frame-shift. ORF1 encodes a shorter isoform with a CCHC-type zinc finger motif containing a sequence characteristic of gag proteins of most retroviruses and some retrotransposons. The longer isoform is the result of -1 translational frame-shifting leading to translation of a gag/pol-like protein combining RF1 and RF2. It contains the active-site consensus sequence of the protease domain of pol proteins. Additional isoforms resulting from alternatively spliced transcript variants, as well as from use of upstream non-AUG (CUG) start codon, have been reported for this gene. PEG10: Prevents apoptosis in hepatocellular carcinoma (HCC) cells through interaction with SIAH1, a mediator of apoptosis. May also have a role in cell growth promotion and hepatoma formation. Inhibits the TGF-beta signaling by interacting with the TGF-beta receptor ALK1. When overexpressed, induces the formation of cellular extension, such as filipodia in association with ALK1. Involved at the immediate early stage of adipocyte differentiation. May bind to the 5 -GCCTGTCTTT-3 DNA sequence of the MB1 domain in the myelin basic protein (MBP) promoter. Up-regulated by MYC and dihydrotestosteron (DHT). Interacts with ALK1, SIAH1 and SIAH2. 2 isoforms of the human protein are produced by ribosomal frameshifting.
