Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for NOTCH3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any NOTCH3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NOTCH3 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NOTCH3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes the third discovered human homologue of the Drosophilia melanogaster type I membrane protein notch. In Drosophilia, notch interaction with its cell-bound ligands (delta, serrate) establishes an intercellular signalling pathway that plays a key role in neural development. Homologues of the notch-ligands have also been identified in human, but precise interactions between these ligands and the human notch homologues remains to be determined. Mutations in NOTCH3 have been identified as the underlying cause of cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL).
