Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for UCMA has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any UCMA present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for UCMA is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of UCMA bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a chondrocyte-specific, highly charged protein that is abundantly expressed in the upper immature zone of fetal and juvenile epiphyseal cartilage. The encoded protein undergoes proteolytic processing to generate a mature protein that is secreted into the extracellular matrix. The glutamic acid residues in the encoded protein undergo gamma carboxylation in a vitamin K-dependent manner. Undercarboxylation of the encoded protein is associated with osteoarthritis in humans. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Jul 2015]
