Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for NLRP1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any NLRP1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NLRP1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NLRP1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a member of the Ced-4 family of apoptosis proteins. Ced-family members contain a caspase recruitment domain (CARD) and are known to be key mediators of programmed cell death. The encoded protein contains a distinct N-terminal pyrin-like motif, which is possibly involved in protein-protein interactions. This protein interacts strongly with caspase 2 and weakly with caspase 9. Overexpression of this gene was demonstrated to induce apoptosis in cells. Multiple alternatively spliced transcript variants encoding distinct isoforms have been found for this gene, but the biological validity of some variants has not been determined.
