Principle of the Assay This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for CHIT1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CHIT1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CHIT1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CHIT1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Chitotriosidase is secreted by activated human macrophages and is markedly elevated in plasma of Gaucher disease patients. The expression of chitotriosidase occurs only at a late stage of differentiation of monocytes to activated macrophages in culture. Human macrophages can synthesize a functional chitotriosidase, a highly conserved enzyme with a strongly regulated expression. This enzyme may play a role in the degradation of chitin-containing pathogens. Several alternatively spliced transcript variants have been described for this gene. [provided by RefSeq, Jan 2012]
