specificity :
This assay has high sensitivity and excellent specificity for detection of human SPARC. No significant cross-reactivity or interference between human SPARC and analogues was observed.
storage stability :
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
other info1 :
Samples: Serum, plasma, tissue homogenates, urine. Assay Type: Sandwich. Detection Range: 62.5 ng/ml - 4000 ng/ml. Sensitivity: The minimum detectable dose of human SPARC is typically less than 15.6 ng/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
products description :
Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for SPARC has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SPARC present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SPARC is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SPARC bound in the initial step. The color development is stopped and the intensity of the color is measured.

ncbi summary :
This gene encodes a cysteine-rich acidic matrix-associated protein. The encoded protein is required for the collagen in bone to become calcified but is also involved in extracellular matrix synthesis and promotion of changes to cell shape. The gene product has been associated with tumor suppression but has also been correlated with metastasis based on changes to cell shape which can promote tumor cell invasion. uniprot summary :
SPARC: Appears to regulate cell growth through interactions with the extracellular matrix and cytokines. Binds calcium and copper, several types of collagen, albumin, thrombospondin, PDGF and cell membranes. There are two calcium binding sites; an acidic domain that binds 5 to 8 Ca(2+) with a low affinity and an EF-hand loop that binds a Ca(2+) ion with a high affinity. Belongs to the SPARC family. Protein type: Secreted; Secreted, signal peptide. Chromosomal Location of Human Ortholog: 5q31.3-q32. Cellular Component: extracellular space; platelet alpha granule membrane; nuclear matrix; cell surface; mitochondrion; cytoplasm; extracellular region; plasma membrane; basement membrane. Molecular Function: collagen binding; protein binding; extracellular matrix binding; calcium ion binding. Biological Process: response to peptide hormone stimulus; platelet activation; response to gravity; receptor-mediated endocytosis; extracellular matrix organization and biogenesis; ossification; response to cAMP; heart development; response to glucocorticoid stimulus; response to lipopolysaccharide; signal transduction; response to L-ascorbic acid; negative regulation of angiogenesis; response to ethanol; platelet degranulation; response to cadmium ion; response to cytokine stimulus; negative regulation of endothelial cell proliferation; response to lead ion; regulation of cell morphogenesis; blood coagulation; response to calcium ion; inner ear development; lung development.
