Priciple of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for CHKB has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CHKB present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CHKB is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CHKB bound in the initial step. The color development is stopped and the intensity of the color is measured.

Choline kinase (CK) and ethanolamine kinase (EK) catalyze the phosphorylation of choline/ethanolamine to phosphocholine/phosphoethanolamine. This is the first enzyme in the biosynthesis of phosphatidylcholine/phosphatidylethanolamine in all animal cells. The highly purified CKs from mammalian sources and their recombinant gene products have been shown to have EK activity also, indicating that both activities reside on the same protein. The choline kinase-like protein encoded by CHKL belongs to the choline/ethanolamine kinase family; however, its exact function is not known.
