. Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for EPX has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any EPX present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for EPX is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of EPX bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene is a member of the peroxidase gene family and is expressed in eosinophils. The encoded precursor protein is processed into covalently attached heavy and light chains to form the mature enzyme, which functions as an oxidant. The enzyme is released at sites of parasitic infection or allergen stimulation to mediate lysis of protozoa or parasitic worms. The gene is found in a cluster of three peroxidase genes at chromosome 17q23. Mutations in this gene result in eosinophil peroxidase deficiency. [provided by RefSeq, Sep 2009]
