Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for PYCARD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PYCARD present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PYCARD is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PYCARD bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes an adaptor protein that is composed of two protein-protein interaction domains: a N-terminal PYRIN-PAAD-DAPIN domain (PYD) and a C-terminal caspase-recruitment domain (CARD). The PYD and CARD domains are members of the six-helix bundle death domain-fold superfamily that mediates assembly of large signaling complexes in the inflammatory and apoptotic signaling pathways via the activation of caspase. In normal cells, this protein is localized to the cytoplasm; however, in cells undergoing apoptosis, it forms ball-like aggregates near the nuclear periphery.
