Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for EGLN1/C1ORF12/PNAS-118/PNAS-137 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any EGLN1/C1ORF12/PNAS-118/PNAS-137 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for EGLN1/C1ORF12/PNAS-118/PNAS-137 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of EGLN1/C1ORF12/PNAS-118/PNAS-137 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Hydroxylates a specific proline found in each of the oxygen-dependent degradation (ODD) domains (N-terminal, NODD, and C-terminal, CODD) of HIF1A. Also hydroxylates HIF2A. Has a preference for the CODD site for both HIF1A and HIF1B. Hydroxylated HIFs are then targeted for proteasomal degradation via the von Hippel-Lindau ubiquitination complex. Under hypoxic conditions, the hydroxylation reaction is attenuated allowing HIFs to escape degradation resulting in their translocation to the nucleus, heterodimerization with HIF1B, and increased expression of hypoxy-inducible genes.
