Samples: For the quantitative determination of human Egl nine homolog 1 (EGLN1/C1orf12/PNAS-118/PNAS-137) concentrations in serum, plasma, tissue homogenates, cell lysates. Detection Range: 31.25 pg/ml -2000 pg/ml.
other info2 :
Sensitivity: The minimum detectable dose of human EGLN1/C1ORF12/PNAS-118/PNAS-137 is typically less than 7.8 pg/ml.The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations. Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
products description :
Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for EGLN1/C1ORF12/PNAS-118/PNAS-137 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any EGLN1/C1ORF12/PNAS-118/PNAS-137 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for EGLN1/C1ORF12/PNAS-118/PNAS-137 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of EGLN1/C1ORF12/PNAS-118/PNAS-137 bound in the initial step. The color development is stopped and the intensity of the color is measured.

ncbi summary :
The protein encoded by this gene catalyzes the post-translational formation of 4-hydroxyproline in hypoxia-inducible factor (HIF) alpha proteins. HIF is a transcriptional complex that plays a central role in mammalian oxygen homeostasis. This protein functions as a cellular oxygen sensor, and under normal oxygen concentration, modification by prolyl hydroxylation is a key regulatory event that targets HIF subunits for proteasomal destruction via the von Hippel-Lindau ubiquitylation complex. Mutations in this gene are associated with erythrocytosis familial type 3 (ECYT3). [provided by RefSeq, Nov 2009]
uniprot summary :
Function: Cellular oxygen sensor that catalyzes, under normoxic conditions, the post-translational formation of 4-hydroxyproline in hypoxia-inducible factor (HIF) alpha proteins. Hydroxylates a specific proline found in each of the oxygen-dependent degradation (ODD) domains (N-terminal, NODD, and C-terminal, CODD) of HIF1A. Also hydroxylates HIF2A. Has a preference for the CODD site for both HIF1A and HIF1B. Hydroxylated HIFs are then targeted for proteasomal degradation via the von Hippel-Lindau ubiquitination complex. Under hypoxic conditions, the hydroxylation reaction is attenuated allowing HIFs to escape degradation resulting in their translocation to the nucleus, heterodimerization with HIF1B, and increased expression of hypoxy-inducible genes. EGLN1 is the most important isozyme under normoxia and, through regulating the stability of HIF1, involved in various hypoxia-influenced processes such as angiogenesis in retinal and cardiac functionality. Target proteins are preferencially recognized via a LXXLAP motif.6 PublicationsManual assertion based on experiment in:Ref.8. Catalytic activity: Hypoxia-inducible factor-L-proline + 2-oxoglutarate + O2 = hypoxia-inducible factor-trans-4-hydroxy-L-proline + succinate + CO2. Cofactor: Binds 1 Fe2+ ion per subunit. Enzyme regulation: Following exposure to hypoxia, activated in HeLa cells but not in cardiovascular cells.2 PublicationsManual assertion based on experiment in:Ref.12
