Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for UMOD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any UMOD present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for UMOD is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of UMOD bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a glycoprotein that is the most abundant protein in mammalian urine under physiological conditions. It is synthesized in the kidney as a glycosyl-phosphatidylinositol anchored protein and released into urine as a soluble form by proteolytic cleavage. It is thought to regulate water and salt balance in the thick ascending limb of Henle and to protect against urinary tract infection and calcium oxalate crystal formation. In mouse deficiency of this gene is associated with increased susceptibility to bacterial infections and formation of calcium crystals in kidneys.
