PTGDS_HUMAN
specificity :
This assay has high sensitivity and excellent specificity for detection of human PTGDS. No significant cross-reactivity or interference between human PTGDS and analogues was observed.
storage stability :
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
other info1 :
Samples: Serum, plasma, cell culture supernates, tissue homogenates. Assay Type: Sandwich. Detection Range: 0.78 ng/ml-50 ng/ml. Sensitivity: The minimum detectable dose of human PTGDS is typically less than 0.2 ng/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
products description :
Priciple of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for PTGDS has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PTGDS present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PTGDS is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PTGDS bound in the initial step. The color development is stopped and the intensity of the color is measured.

ncbi summary :
The protein encoded by this gene is a glutathione-independent prostaglandin D synthase that catalyzes the conversion of prostaglandin H2 (PGH2) to postaglandin D2 (PGD2). PGD2 functions as a neuromodulator as well as a trophic factor in the central nervous system. PGD2 is also involved in smooth muscle contraction/relaxation and is a potent inhibitor of platelet aggregation. This gene is preferentially expressed in brain. Studies with transgenic mice overexpressing this gene suggest that this gene may be also involved in the regulation of non-rapid eye movement sleep. [provided by RefSeq, Jul 2008]
uniprot summary :
PTGDS: Catalyzes the conversion of PGH2 to PGD2, a prostaglandin involved in smooth muscle contraction/relaxation and a potent inhibitor of platelet aggregation. Involved in a variety of CNS functions, such as sedation, NREM sleep and PGE2-induced allodynia, and may have an anti-apoptotic role in oligodendrocytes. Binds small non-substrate lipophilic molecules, including biliverdin, bilirubin, retinal, retinoic acid and thyroid hormone, and may act as a scavenger for harmful hydrophopic molecules and as a secretory retinoid and thyroid hormone transporter. Possibly involved in development and maintenance of the blood-brain, blood-retina, blood-aqueous humor and blood-testis barrier. It is likely to play important roles in both maturation and maintenance of the central nervous system and male reproductive system. Belongs to the calycin superfamily. Lipocalin family. Protein type: Secreted, signal peptide; EC 5.3.99.2; Endoplasmic reticulum; Isomerase; Lipid Metabolism - arachidonic acid; Secreted. Chromosomal Location of Human Ortholog: 9q34.2-q34.3. Cellular Component: Golgi apparatus; extracellular space; endoplasmic reticulum membrane; nuclear membrane; rough endoplasmic reticulum; perinuclear region of cytoplasm; extracellular region. Molecular Function: retinoid binding; prostaglandin-D synthase activity; transporter activity; fatty acid binding. Biological Process: transport; cyclooxygenase pathway; response to glucocorticoid stimulus; arachidonic acid metabolic process; prostaglandin biosynthetic process; regulation of circadian sleep/wake cycle, sleep
