This assay has high sensitivity and excellent specificity for detection of Human SRD5A1. No significant cross-reactivity or interference between Human SRD5A1 and analogues was observed.

Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV% is less than 8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision (Precision between assays): CV% is less than 10%. Three samples of known concentration were tested in twenty assays to assess. Sample Type: serum, plasma, tissue homogenates and cell lysates. Detection Range: 0.156 ng/ml-10 ng/ml. Detection Wavelength: 450 nm. Sensitivity: 0.039 ng/ml

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for SRD5A1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SRD5A1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SRD5A1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SRD5A1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Steroid 5-alpha-reductase (EC 1.3.99.5) catalyzes the conversion of testosterone into the more potent androgen, dihydrotestosterone (DHT). Also see SRD5A2 (MIM 607306).[supplied by OMIM, Mar 2008]

Function: Converts testosterone into 5-alpha-dihydrotestosterone and progesterone or corticosterone into their corresponding 5-alpha-3-oxosteroids. It plays a central role in sexual differentiation and androgen physiology. Catalytic activity: A 3-oxo-5-alpha-steroid + NADP+ = a 3-oxo-Delta(4)-steroid + NADPH.
