Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV% is less than 8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision (Precision between assays): CV% is less than 10%. Three samples of known concentration were tested in twenty assays to assess. Detection Wavelength: 450 nm. Sample Volume: 50-100ul
products description :
Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for pantetheinase has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any pantetheinase present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for pantetheinase is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of pantetheinase bound in the initial step. The color development is stopped and the intensity of the color is measured.

ncbi summary :
This gene encodes a member of the vanin family of proteins, which share extensive sequence similarity with each other, and also with biotinidase. The family includes secreted and membrane-associated proteins, a few of which have been reported to participate in hematopoietic cell trafficking. No biotinidase activity has been demonstrated for any of the vanin proteins, however, they possess pantetheinase activity, which may play a role in oxidative-stress response. This protein, like its mouse homolog, is likely a GPI-anchored cell surface molecule. The mouse protein is expressed by the perivascular thymic stromal cells and regulates migration of T-cell progenitors to the thymus. This gene lies in close proximity to, and in the same transcriptional orientation as, two other vanin genes on chromosome 6q23-q24. [provided by RefSeq, Feb 2009]
uniprot summary :
VNN1: Amidohydrolase that hydrolyzes specifically one of the carboamide linkages in D-pantetheine thus recycling pantothenic acid (vitamin B5) and releasing cysteamine. Belongs to the CN hydrolase family. BTD/VNN subfamily. Protein type: Hydrolase; Motility/polarity/chemotaxis; Membrane protein, GPI anchor; Cell adhesion; Membrane protein, integral; Cofactor and Vitamin Metabolism - pantothenate and CoA biosynthesis; EC 3.5.1.92. Chromosomal Location of Human Ortholog: 6q23-q24. Cellular Component: plasma membrane; integral to membrane. Molecular Function: pantetheine hydrolase activity. Biological Process: positive regulation of T cell differentiation in the thymus; chronic inflammatory response; cell-cell adhesion; pantothenate metabolic process; innate immune response; response to oxidative stress; inflammatory response; cell motility; acute inflammatory response
