specificity :
This assay has high sensitivity and excellent specificity for detection of human BRAF. No significant cross-reactivity or interference between human BRAF and analogues was observed.
storage stability :
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
other info1 :
Samples: Serum, plasma, Cell lysates. Assay Type: Sandwich. Detection Range: 31.25 pg/ml -2000 pg/ml. Sensitivity: The minimum detectable dose of human BRAF is typically less than 7.81 pg/ml.The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
products description :
Princple of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for BRAF has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any BRAF present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for BRAF is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of BRAF bound in the initial step. The color development is stopped and the intensity of the color is measured.

ncbi summary :
This gene encodes a protein belonging to the raf/mil family of serine/threonine protein kinases. This protein plays a role in regulating the MAP kinase/ERKs signaling pathway, which affects cell division, differentiation, and secretion. Mutations in this gene are associated with cardiofaciocutaneous syndrome, a disease characterized by heart defects, mental retardation and a distinctive facial appearance. Mutations in this gene have also been associated with various cancers, including non-Hodgkin lymphoma, colorectal cancer, malignant melanoma, thyroid carcinoma, non-small cell lung carcinoma, and adenocarcinoma of lung. A pseudogene, which is located on chromosome X, has been identified for this gene. [provided by RefSeq, Jul 2008]
uniprot summary :
BRAF: a tyrosine kinase-like kinase of the RAF family. Involved in the transduction of mitogenic signals from the cell membrane to the nucleus. May play a role in the postsynaptic responses of hippocampal neuron. Frequently mutated in thyroid cancers, skin melanomas and at lower frequency in a wide range of human cancers. An activating mutation, mimicking phosphorylation of the activation loop, is seen in 60% of malignant melanoma samples. Raf mutations are generally exclusive to Ras activating mutations. Activating mutations are also seen in ~10% of colorectal cancers, in lung cancers and gliomas, and at a lower rate in several other tumors. Inactivating mutations are also seen and may result in activation of c-Raf and Erk. Mutations in B-Raf, MEK1 and MEK2 also associated with cardiofaciocutaneous syndrome, displaying morphological, cardiac and mental defects. Approved Inhibitor: Nexavar/Sorafenib. Protein type: EC 2.7.11.1; Kinase, protein; Protein kinase, TKL; Protein kinase, Ser/Thr (non-receptor); Oncoprotein; TKL group; RAF family. Chromosomal Location of Human Ortholog: 7q34. Cellular Component: neuron projection; mitochondrion; plasma membrane; nucleus; cytosol. Molecular Function: small GTPase binding; identical protein binding; protein serine/threonine kinase activity; protein binding; MAP kinase kinase kinase activity; protein heterodimerization activity; mitogen-activated protein kinase kinase binding; calcium ion binding; ATP binding; protein kinase activity. Biological Process: response to peptide hormone stimulus; positive T cell selection; response to cAMP; fibroblast growth factor receptor signaling pathway; nerve growth factor receptor signaling pathway; activation of MAPKK activity; CD4-positive, alpha beta T cell differentiation; protein heterooligomerization; somatic stem cell maintenance; MAPKKK cascade; protein amino acid phosphorylation; positive regulation of peptidyl-serine phosphorylation; regulation of cell proliferation; synaptic transmission; organ morphogenesis; myeloid progenitor cell differentiation; small GTPase mediated signal transduction; positive regulation of stress fiber formation; visual learning; negative regulation of neuron apoptosis; negative regulation of apoptosis.
