Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for SPTAN1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SPTAN1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SPTAN1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SPTAN1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Spectrins are a family of filamentous cytoskeletal proteins that function as essential scaffold proteins that stabilize the plasma membrane and organize intracellular organelles. Spectrins are composed of alpha and beta dimers that associate to form tetramers linked in a head-to-head arrangement. This gene encodes an alpha spectrin that is specifically expressed in nonerythrocytic cells. The encoded protein has been implicated in other cellular functions including DNA repair and cell cycle regulation. Mutations in this gene are the cause of early infantile epileptic encephalopathy-5.
