The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest mouse FSTL3 concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.

Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for FSTL3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells with a Horseradish Peroxidase (HRP) conjugated antibody specific for FSTL3. Following a wash to remove any unbound reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FSTL3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Inhibits activin A-, activin B-, BMP2- and MSDT-induced cellular signaling; more effective on activin A than on activin B. Involved in bone formation; inhibits osteoclast differentiationc. Involved in hematopoiesis; involved in differentiation of hemopoietic progenitor cells, increases hematopoietic cell adhesion to fibronectin and seems to contribute to the adhesion of hematopoietic precursor cells to the bone marrow stroma. Isoform 2 or the nuclear form is probably involved in transcriptional regulation via interaction with MLLT10. A chromosomal aberration involving FSTL3 is found in a case of B-cell chronic lymphocytic leukemia.
