Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for KRT1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any KRT1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for KRT1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of KRT1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is a member of the keratin gene family. The type II cytokeratins consist of basic or neutral proteins which are arranged in pairs of heterotypic keratin chains coexpressed during differentiation of simple and stratified epithelial tissues. This type II cytokeratin is specifically expressed in the spinous and granular layers of the epidermis with family member KRT10 and mutations in these genes have been associated with bullous congenital ichthyosiform erythroderma. The type II cytokeratins are clustered in a region of chromosome 12q12-q13. [provided by RefSeq, Jul 2008]
