Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ABL1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ABL1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ABL1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ABL1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Acetyl-CoA carboxylase (ACC) is a complex multifunctional enzyme system. ACC is a biotin-containing enzyme which catalyzes the carboxylation of acetyl-CoA to malonyl-CoA, the rate-limiting step in fatty acid synthesis. There are two ACC forms, alpha and beta, encoded by two different genes. ACC-alpha is highly enriched in lipogenic tissues. The enzyme is under long term control at the transcriptional and translational levels and under short term regulation by the phosphorylation/dephosphorylation of targeted serine residues and by allosteric transformation by citrate or palmitoyl-CoA.
