specificity :
This assay has high sensitivity and excellent specificity for detection of rat COMT. No significant cross-reactivity or interference between rat COMT and analogues was observed.
storage stability :
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
other info1 :
Samples: Serum, plasma, tissue homogenates, Cell lysates. Assay Type: Sandwich. Detection Range: 31.25 pg/ml -2000 pg/ml. Sensitivity: The minimum detectable dose of rat COMT is typically less than 7.8 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
products description :
Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for COMT has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any COMT present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for COMT is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of COMT bound in the initial step. The color development is stopped and the intensity of the color is measured.

catalyzes the conversion of S-adenosyl-L-methionine and catechol to S-adenosyl-L-homocysteine and guaiacol [RGD, Feb 2006]
uniprot summary :
COMT: Catalyzes the O-methylation, and thereby the inactivation, of catecholamine neurotransmitters and catechol hormones. Also shortens the biological half-lives of certain neuroactive drugs, like L-DOPA, alpha-methyl DOPA and isoproterenol. Belongs to the mammalian catechol-O-methyltransferase family. 2 isoforms of the human protein are produced by alternative initiation. Protein type: EC 2.1.1.6; Membrane protein, integral; Methyltransferase; Amino Acid Metabolism - tyrosine. Cellular Component: postsynaptic membrane; mitochondrion; membrane; axon; dendrite; dendritic spine; plasma membrane; integral to membrane; cytosol. Molecular Function: magnesium ion binding; catechol O-methyltransferase activity. Biological Process: response to drug; methylation; estrogen metabolic process; cellular response to phosphate starvation; neurotransmitter catabolic process; dopamine catabolic process; negative regulation of smooth muscle cell proliferation; short-term memory; S-adenosylmethionine metabolic process; response to pain; S-adenosylhomocysteine metabolic process; response to lipopolysaccharide; learning; female pregnancy; reproductive process in a multicellular organism; catechol metabolic process; response to organic cyclic substance; positive regulation of homocysteine metabolic process; dopamine metabolic process; negative regulation of dopamine metabolic process; regulation of sensory perception of pain; developmental process
