Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for sCD276/sB7-H3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any sCD276/sB7-H3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for sCD276/sB7-H3 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of sCD276/sB7-H3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene belongs to the immunoglobulin superfamily, and thought to participate in the regulation of T-cell-mediated immune response. Studies show that while the transcript of this gene is ubiquitously expressed in normal tissues and solid tumors, the protein is preferentially expressed only in tumor tissues. Additionally, it was observed that the 3' UTR of this transcript contains a target site for miR29 microRNA, and there is an inverse correlation between the expression of this protein and miR29 levels, suggesting regulation of expression of this gene product by miR29.
