Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ANPEP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ANPEP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ANPEP is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ANPEP bound in the initial step. The color development is stopped and the intensity of the color is measured.

Function: Broad specificity aminopeptidase. Plays a role in the final digestion of peptides generated from hydrolysis of proteins by gastric and pancreatic proteases. May be involved in the metabolism of regulatory peptides of diverse cell types, responsible for the processing of peptide hormones, such as angiotensin III and IV, neuropeptides, and chemokines and involved the cleavage of peptides bound to major histocompatibility complex class II molecules of antigen presenting cells. May have a role in angiogenesis . Catalytic activity: Release of an N-terminal amino acid, Xaa- -Yaa- from a peptide, amide or arylamide. Xaa is preferably Ala, but may be most amino acids including Pro (slow action). When a terminal hydrophobic residue is followed by a prolyl residue, the two may be released as an intact Xaa-Pro dipeptide. Cofactor: Binds 1 zinc ion per subunit
