Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for PADI2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PADI2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PADI2 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PADI2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a member of the peptidyl arginine deiminase family of enzymes, which catalyze the post-translational deimination of proteins by converting arginine residues into citrullines in the presence of calcium ions. The family members have distinct substrate specificities and tissue-specific expression patterns. The type II enzyme is the most widely expressed family member. Known substrates for this enzyme include myelin basic protein in the central nervous system and vimentin in skeletal muscle and macrophages. This enzyme is thought to play a role in the onset and progression of neurodegenerative human disorders, including Alzheimer disease and multiple sclerosis, and it has also been implicated in glaucoma pathogenesis.
