Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for WNT4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any WNT4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for WNT4 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of WNT4 bound in the initial step. The color development is stopped and the intensity of the color is measured.

May be a signaling molecule which affects the development of discrete regions of tissues. Is likely to signal over only few cell diameters. Overexpression may be associated with abnormal proliferation in human breast tissue. Defects in WNT4 are a cause of Rokitansky-Kuster-Hauser syndrome (RKH syndrome); also called Mayer- Rokitansky-Kuster-Hauser syndrome (MRKH syndrome or MRKH anomaly). RKH syndrome is characterized by utero-vaginal atresia in otherwise phenotypically normal female with a normal 46,XX karyotype.
