Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for CD59 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CD59 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CD59 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CD59 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a cell surface glycoprotein that regulates complement-mediated cell lysis, and it is involved in lymphocyte signal transduction. This protein is a potent inhibitor of the complement membrane attack complex, whereby it binds complement C8 and/or C9 during the assembly of this complex, thereby inhibiting the incorporation of multiple copies of C9 into the complex, which is necessary for osmolytic pore formation. This protein also plays a role in signal transduction pathways in the activation of T cells. Mutations in this gene cause CD59 deficiency, a disease resulting in hemolytic anemia and thrombosis, and which causes cerebral infarction.
