Activity (Specifications/test method): GSK-3alpha fusion protein at a concentration of 2 ug/20 ul reaction was phosphorylated in an in vitro kinase assay with 200 uM ATP. After 30 minutes incubation at 30 degree C, phosphorylation was detected by Western blot with phospho-specific GSK-3alpha antibodies. Handling: Centrifuge the vial prior to opening.

Background: GSK-3 is one of the Akt substrates. It can be phosphorylated by Akt at Ser21 of GSK-3alpha or Ser9 of GSK-3beta. The phosphorylation of GSK negatively regulates its kinase activity. The recombinant GSK-3alpha fusion protein serves as a useful substrate for assaying Akt kinase activity.

GSK3 phophorylates tau, the principal component of neurofibrillary tangles in Alzheimer disease and is required for maximal production of amyloid plaque peptides by secretase. A GSK3 promoter SNP effects progression of bipolar disorder. The GSK3 inhibitor, lithium, is used to treat bipolar disorder and is seen to block plaque formation.
