Principle of the Assay: The MyBioSource' Mouse CD25 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse CD25 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Mouse CD25 coated on a 96-well plate. Standards and samples are pipetted into the wells and CD25 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse CD25 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD25 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm!!Background/Introduction: CD25 is the alpha chain of the IL-2 receptor. It is a type I transmembrane protein present on activated T cells, activated B cells, some thymocytes, myeloid precursors, and oligodendrocytes that associates with CD122 to form a heterodimer that can act as a high-affinity receptor for IL-2. CD25 has been used as a marker to identify CD4+FoxP3+ regulatory T cells in mice. By in situ hybridization, the IL2R gene was mapped to 10p15-p14. Its soluble form, called sIL-2R may be elevated in these diseases and is occasionally used to track disease progression.
